چکیده

We report on development of a microfluidic device and optical system for measurement of occlusive platelet thrombosis at initial shear rates ranging from physiological to pathological conditions from (500 – 13000 s) within four stenotic channels in unfractionated blood. Real-time, label-free measurement of platelet thrombosis was performed by measuring optical radiation transmitted transversely through flow channels. Occlusive thrombosis occurred at shear rates above 4000 s and measurements showed average differences in of 8.4% and 3.97% compared with traditional flow-rate and white-light microscopy methods, respectively.

Microfluidic system for simultaneous optical measurement of platelet aggregation at multiple shear rates in whole blood.

Thrombosis is the pathological formation of platelet aggregates which occlude blood flow causing stroke and heart attack-the leading causes of death in developed nations. Instrumentation for diagnosing and exploring treatments for pathological platelet aggregation thus has the potential for major clinical impact. Most current thrombosis methods focus on single flow conditions, non-occlusive pla...

A simple method for activating the platelets used in microfluidic platelet aggregation tests: Stirring-induced platelet activation

High-shear stimulation is well known as one of the key factors affecting platelet activation and aggregation, which can lead to the formation of a thrombus. In one of our previous studies, we introduced migration distance-based platelet function analysis in a microfluidic system. In this study, we set out to examine the effects of stirring on shear-induced platelet activation and aggregation in...

Relative Contributions of von Willebrand Factor and Platelets in High Shear Thrombosis

Platelets and von Willebrand factor (VWF) are recognized as important mediators of thrombosis at high shear rates, but their relative contributions are unclear. We employ a stenotic microfluidic test system to induce thrombus formation to occlusion on collagen at pathologic shear rates of 3500-6000 s-1. To obtain blood analogs with reduced platelet and VWF concentrations, human whole blood was ...

Microfluidic Thrombosis under Multiple Shear Rates and Antiplatelet Therapy Doses

The mainstay of treatment for thrombosis, the formation of occlusive platelet aggregates that often lead to heart attack and stroke, is antiplatelet therapy. Antiplatelet therapy dosing and resistance are poorly understood, leading to potential incorrect and ineffective dosing. Shear rate is also suspected to play a major role in thrombosis, but instrumentation to measure its influence has been...

An Overview on Platelet-derived Microparticles in Platelet Concentrates: blood collection, method preparation and storage

Preparations of platelet concentrates (PCs) that are stored under blood bank conditions and used for transfusion purposes, appear to be enriched in platelet derived-microparticles (PMPs) with high coagulant activity that may change platelet efficacy and safety issues. High shear stress could cause shedding of PMPs from the platelet plasma membrane, platelet aggregation, and activation of the co...
